It is originally used as an explosive, a pesticide, as well as a range of other industrial processes including- dye, wood preserver, herbicide and photographic developer. This causes the mitochondria to use more energy to create ATP, thus, increasing the metabolic rate and increasing caloric expenditure which leads to weight loss 4.
In recent years, there has been a resurgence of DNP use, largely in the bodybuilding community. DNP can assist in weight loss. This is the main reported benefit of DNP usage, and it does this through several associated mechanisms listed below.
DNP increases the metabolic rate by stimulating the uncoupling of oxidative phosphorylation This causes the cell to undergo more ATP production to meet energy demands. Since the production of ATP actually costs energy, this results in a greater metabolic rate, thereby leading to weight loss DNP also causes an increase in internal heat production By altering the electrochemical gradient of protons in favor of producing less ATP, the result is more energy being lost as heat production instead of conversion to ATP This increase in heat production leads to an increase in caloric consumption, which again leads to weight loss DNP causes an upregulation in glycolysis Researchers found that the combination of the stimulation of glycolysis and the inhibition of uncoupling resulted in a large increase in carbohydrate depletion 16 All of these mechanisms serve the same purpose: to decrease the efficiency of energy production as a means of increasing energy expenditure.
However, these mechanisms greatly disturb normal cell function and are all potentially lethal DNP may have promising effects on age-related neurological disorders according to recent research. The research is preliminary and has not yet been tested in humans, but it certainly warrants further examination. One study used rat models to assess the effects of DNP use after suffering an ischemic stroke.
DNP was shown to reduce impact volume thereby reducing the damage done by the stroke This is, of course, an important finding that may have benefits in reducing impairment after suffering a stroke. In all three scenarios, there is existing mitochondrial dysfunction, which the DNP appears to regulate 2425 Also, the researchers are not exactly sure of the mechanism by which DNP is acting to regulate the mitochondrial function.Girl Dies After Diet Pill Overdose "Burned Her From Within"
It is also important to note that these studies all used rat models and are one-of-kind, so much further research is needed to determine the safety and efficacy of DNP use in the treatment of certain neurological disorders.
DNP makes dieting somewhat effortless. DNP decreases carb tolerance. Low carb diets have shown time and time again to be an effective weight loss agent. DNP causes an inability to handle large amounts of carbohydrates and often forces users to go lower carb.
This often results in even more weight loss. DNP can combat liver disease. Unfortunately, there are no approved drugs for treating this condition.By Charlie Moore For Mailonline. The mother of a year-old woman who died after taking illegal diet pills has criticised the hospital where she was treated for 'doing nothing'. Beth Shipsey told paramedics she ate several tablets containing industrial fat-burning chemical DNP at home in February. She was rushed to Worcestershire Royal Hospital where her parents pleaded with nurses to treat her but there were delays recognising her condition, a report handed to the family has found.
Beth's father Doug said she was taken to a resuscitation room but was later moved to make way for more seriously ill patients. Her mother Carole, who has been a nurse for 39 years, said she told staff that her daughter was having a respiratory arrest but a tracheotomy was performed too late.
Beth died just days after the Trust was told by the Quality Care Commission to 'make significant improvements'.
'Overwhelmed' hospital in Worcester missed DNP overdose
She was a victim of rape and suffered mental health difficulties. Recalling her daughter's final day with quivering voice, Mrs Shipsey told the BBC : 'Beth looked at me and just called my name. That was the last time she actually spoke and she just called "mum, mum, mum". Beth Shipsey, from Worcestershire, told paramedics she ate several tablets containing industrial fat-burning chemical DNP at home in February.
And she alerted somebody on the internet that she took them. Talking about the hospital service, she said: 'I'm struggling to even believe it myself that they did nothing. In an internal report, Worcestershire Acute Trust admitted delays in recognising Beth's condition but said it believed her death was inevitable. It said: 'An overwhelmed department led to a delay in recognition of a potentially fatal overdose and delayed implementation of cooling measures.
DNP is sold as a weight loss aid, but has been described as 'extremely dangerous to human health' by doctors. It is sold mostly over the internet under a number of different names but contains 2, 4-Dinitrophenol. It is marketed mainly to bodybuilders as a weight loss aid as it is thought to dramatically boost metabolism. The manufactured drug is yellow and odourless and was previously used as a herbicide and fungicide.
It was launched as a slimming aid in the U. Depending on the amount consumed, signs of acute poisoning could include nausea, vomiting, restlessness, flushed skin, sweating, dizziness, headaches, rapid respiration and irregular heart-beat, possibly leading to coma and death. Worcestershire Acute Trust admitted delays in recognising Beth's condition but said it believed her death was inevitable.
The results of this have been shared with the family. Mr Shipsey believes his daughter took the pills as a 'cry for help' rather than to end her life. An inquest is due to be held in January Eloise Parry burned up from the inside, dying in agony three hours after taking DNP in a misguided attempt to control her weight.
Eloise - known as Ella - 21, from Shrewsbury, who suffered from the eating disorder bulimia, drove herself to hospital after taking eight 'fat burner' pills which contained DNP. The fatal dose is just two. Medical staff told her there was no antidote to DNP poisoning. She overheated and died three hours later.

Then DNP was discovered for human consumption that led to Weight loss and it became a popular drug. It was used as an over the counter medication and was sold without a description to the public. Stanford clinics prescribed 1. Dan Duchaine made DNP popular who discovered this in prison initially. Then there was a Russian doctor called Dr. Russian soldiers used DNP to keep warm during the rough winters.
DNP is still illegal however is it available to purchase online even though it was associated with deaths. The initial reports showed DNP increased metabolic rates reducing fat loss. By Oxidation directly on the tissues and cells, DNP promoted fat loss. When fast foods and refined carbs came on America and just a general emphasis on sugar and processed food was on the rise.
This can Obviously be highly dangerous. I am not promoting this product I am simply adding research. These websites additionally describe the potential toxicity associated with the use of DNP including the potential for hypothermia and death. They recommend that should body temperature rise above It has shown Liver damage but it is yet to be fully proven on how that process works. Kidney damage is also undiscovered. In the initial blood testing, there were possibilities that DNP can affect the blood panel.
I do NOT recommend this substance, please consult a medical specialist for experimentation, this is for research purposes only. Clearly, discoveries still need to be made in clinical trials with DNP in terms of side effects. Fatal effects included within its initial studies, Within large dosages, with the drug stimulated heat production that may increase with up to percent. This is clearly unheard of compared to other similar fat-burning chemicals.
The first fatal poisoning was a physician who took DNP for therapeutic purposes. More studies showed skin reactions they showed swelling of the skin. The second death was related to a female who took the drug at their own responsibility.Last week, an inquest into his death found that it was caused by a weight-loss supplement containing 2,4-Dinitrophenol, also known as DNP.
This illegal drug is marketed as a miracle weight-loss aid and is often targeted towards the bodybuilding community. But the rapid weight loss comes at a very high price — as demonstrated not only by Willis' death, but by the deaths of young men and women just like him.
DNP rapidly accelerates one's basal metabolic rate, which can lead to quick weight loss. But it actually was never created to be used as a weight loss aid. It was used by the French for manufacturing munitions during World War I. It's also been used as a pesticide, as a dye, and as a wood preserver. But inwhen DNP was first tried for human consumption, Stanford scientists discovered the bright-yellow compound, manufactured in both powder and crystal form, had fat-shredding properties.
DNP was then marketed as a miracle over-the-counter weight-loss drug — that is, until the adverse affects of the drug began to surface, ranging from dehydration and cataracts to, most frighteningly, death.
Essentially, it boosts basal metabolic rate to accelerate fat burn, resulting in high internal body temperature as the dose increases. These long-term effects were noted back in the s. But in recent years, it's been making a resurgence as a weight-loss drug— thanks in part to a physician named Dr.
Nicholas Bachynsky, who, in the 's, began treating people with DNPre-labeling it "Mitcal," and falsely claimed it to have a multitude of health benefits in addition to weight loss. He was later imprisoned for fraud— but the ramifications of his actions are still being felt today. Although DNP is banned in both the United States and the United Kingdom, it is still easily available for purchase online— and it's being marketed towards bodybuilders as an easy way to drop body fat, fast.
With access to the drug now easier than ever, there has been a marked increase in the number of DNP-attributed deaths since the turn of the centuryespecially among fit, young people. A case study from examined the case of a year-old man who died as a result of taking DNP.
In a casea teenage girl also died after purchasing DNP on the internet as a weight-loss supplement. Both people had purchased DNP online thinking it was a "safe" weight-loss aid, despite a nearly year rule that has deemed DNP highly dangerous and unfit for human consumption. While it can be tempting to buy a pill that claims to make your fat disappearknow that it's indeed too good to be true.
Your body will thank you for taking a much more practical and safe approach to long-term weight loss— eating right and exercising regularly.

Editor—There are only a few cases in the literature of 2,4-dinitrophenol DNP poisoning in adults resulting in death. The prevalence of non-prescription weight loss measures is increasing.
DNP has also been used as a herbicide and also as a photographic developing chemical. Its use has now resurfaced via the internet. A yr-old lady was admitted to accident and emergency complaining of fatigue, nausea, and excessive sweating. She admitted to starting a new diet tablet bought over the internet a week before her admission. She had doubled the recommended dose for faster results.

DNP causes a hyper-metabolic state by uncoupling oxidative phosphorylation. Energy is released in the mitochondria as heat.
The body attempts to compensate by gluconeogenesis, glycolysis, and lipolysis. Toxic doses will result in uncontrolled thermogenesis leading to hyperthermia and systemic responses to elevated body temperature. Renal, hepatic, and neurological sequelae can occur. In cases of severe toxicity, death may be followed by prompt rigor mortis. Management involves aggressive supportive measures and use of benzodiazepines, but because of a large volume of distribution, DNP is not amenable to dialysis or haemoperfusion.
Raised free intracellular calcium causes muscle contraction and hyperthermia. Successful use of dantrolene has been described. This is thought to help to facilitate heat dissipation in uncontrolled DNP-related hyperthermia. We stress importance of early aggressive supportive care and level 2 involvement in cases of DNP poisoning.
We also propose the early use of dantrolene. Google Scholar. It causes uncoupling of oxidative phosphorylation; the classic symptom complex associated with toxicity of phenol-based products such as DNP is a combination of hyperthermia, tachycardia, diaphoresis and tachypnoea, eventually leading to death.
Fatalities related to exposure to DNP have been reported since the turn of the twentieth century.
Role of dantrolene in dinitrophenol (DNP) overdose: A continuing question?
To date, there have been 62 published deaths in the medical literature attributed to DNP. In this review, we will describe the pattern and pathophysiology of DNP toxicity and summarise the previous fatalities associated with exposure to DNP.
The pharmacologic treatment of obesity has been challenging. Previously, amphetamine derivatives, such as dexflenfluramine, fenfluramine and phentermine, were used as centrally active appetite suppressants; however, their use is associated with valvular heart disease and the development of pulmonary hypertension. Regular use of 2,4-dinitrophenol DNP is reported to cause rapid loss of weight, but unfortunately is associated with an unacceptably high rate of significant side effects [ 2 ].
Currently marketing of DNP, which is predominately through the internet, is targeted towards primarily body builders who are attempting to lose fat but retain muscle bulk. There are many regimes for taking the drug, all of which rely on the metabolic stimulatory effects of the drug [ 45 ]. It has a small therapeutic index and is extremely dangerous in overdose. In this review, we will summarise the pharmacology of DNP, the potential mechanisms for its toxicity and the clinical evidence of harm associated with the use of DNP.
All the relevant abstracts were screened by one author JG and those that did not describe death s due to DNP were excluded from the pool of results. All the remaining articles were retrieved and scrutinized to ensure they convincingly described death s due to DNP. All articles describing deaths due to other compounds similar to DNP e.
Citations and references of the remaining articles were searched, retrieved and evaluated in the same way as the original search results. Since then, it has also been used as a dye, wood preserver, herbicide and photographic developer.
It was Maurice Tainter at Stanford University in who discovered that the human consumption of DNP led to significant weight loss and soon it was popularised as a weight loss drug [ 8 ]. It was included in over-the-counter medications and was sold to the public without requiring a prescription. Its use for those wishing to lose weight was encouraged by reports of rapid, safe weight loss [ 78 ].
DNP can cause a significant increase in the basal metabolic rate [ 78 ]. This leads to weight loss by burning more fat and carbohydrates [ 9 ], and weight loss of up to 1. Aftermedical prescription of DNP stopped and cases of poisoning due to medical intake were no longer reported, but case reports of deaths associated with the ingestion of DNP still emerged [ 1415 ].
It is anecdotally reported to have been prescribed to the Russian soldiers during World War II to keep them warm [ 16 ]. Ina physician Dr. It is alleged in subsequent court proceedings that over 14, people were treated by Dr. Individuals using Mitcal started reporting adverse effects, such as fever, shortness of breath and sweating, to the US Food and Drugs Administration in late Following further investigation, Dr.
Bachynsky was convicted in of drug law violations, fined and prohibited from dispensing DNP to any patients. This warning was aimed specifically at bodybuilders, to avoid its use due to significant potential for short-term and long-term harm, following the hospitalisation of a Finish bodybuilder after having taken DNP [ 18 ]. Despite this, DNP remains widely available and as discussed below in more detail, can be purchased over the internet, particularly from online pharmacies.
Additionally, there are instructions for the synthesis of DNP online for self-manufacture. Despite this legislation and warnings of harm associated with its use, reports of deaths due to the use of DNP have increased in the last few years Fig. Summary of previously published fatalities relating to exposure to DNP including basic demographics, amount of exposure and maximal temperature recorded pre-death.
The chemical is a yellow crystalline powder that has a sweet, musty odour and is soluble in water. The dose of DNP per capsule varies from website to website but it is most commonly sold as either or mg capsules. Websites often offer some advice for users around the use of DNP, although this is often targeted towards bodybuilders rather than for generic weight loss.
These websites additionally describe the potential toxicity associated with the use of DNP including the potential for hyperthermia and death. It is a yellow, crystalline solid that has a sweet, musty odor.
It sublimes, is volatile with steam, and is soluble in most organic solvents as well as aqueous alkaline solutions. Its use in high doses as a dieting aid has been identified with severe side-effects, including a number of deaths.Spazio, per parmitano 7 esperimenti italiani dellasi
Commercially, DNP is used as an antiseptic and as a non-selective bioaccumulating pesticide. DNP is particularly useful as a herbicide alongside other closely related dinitrophenol herbicides like 2,4-dinitro- o -cresol DNOCdinoseb and dinoterb.
It is a chemical intermediate in the production of sulfur dyes wood preservatives and picric acid. Although DNP is widely considered too dangerous for clinical use, its mechanism of action remains under investigation as a potential approach for treating obesity. In living cells, DNP acts as a proton ionophorean agent that can shuttle protons hydrogen cations across biological membranes. It dissipates the proton gradient across mitochondria and chloroplast membranes, collapsing the proton motive force that the cell uses to produce most of its ATP chemical energy.
Instead of producing ATP, the energy of the proton gradient is lost as heat. DNP is often used in biochemistry research to help explore the bioenergetics of chemiosmotic and other membrane transport processes.
DNP acts as a protonophoreallowing protons to leak across the inner mitochondrial membrane and thus bypass ATP synthase. This makes ATP energy production less efficient. In effect, part of the energy that is normally produced from cellular respiration is wasted as heat. The inefficiency is proportional to the dose of DNP that is taken.
As the dose increases and energy production is made more inefficient, metabolic rate increases and more fat is burned in order to compensate for the inefficiency and to meet energy demands.
DNP is probably the best known agent for uncoupling oxidative phosphorylation. This leads to free intracellular calcium and causes muscle contraction and hyperthermia. Dantrolene inhibits calcium release from the sarcoplasmic reticulum which reduces intracellular calcium. The resulting muscle relaxation allows heat dissipation. There is little risk to dantrolene administration. Since dantrolene may be effective in reducing hyperthermia caused by agents that inhibit oxidative phosphorylation, early administration may improve outcome.
Information about pharmacokinetics of DNP in humans is limited. DNP is considered to have high acute toxicity. This puts DNP close to cyanide in terms of its toxicity.
Inan incident occurred in a Chinese chemical factory and 20 persons suffered acute DNP poisoning. The factor that limits ever-increasing doses of DNP is not a lack of ATP energy production, but rather an excessive rise in body temperature due to the heat produced during uncoupling. Accordingly, DNP overdose will cause fatal hyperthermiawith body temperature rising to as high as The United Kingdom's Food Standards Agency identifies DNP as "an industrial chemical known to have serious short-term and long-term effects, which can be extremely dangerous to human health.
